Compartmentalization of telomeres through DNA-scaffolded phase separation

نویسندگان

چکیده

Telomeres form unique nuclear compartments that prevent degradation and fusion of chromosome ends by recruiting shelterin proteins excluding DNA damage repair factors. To understand how these dynamic components protect ends, we reconstitute human in vitro show multicomponent liquid condensates with selective biomolecular partitioning on telomeric DNA. TRF1 TRF2 subunits drive phase separation, their N-terminal domains specify interactions vitro.

Scaffolded DNA origami of a DNA tetrahedron molecular container.

We describe a strategy of scaffolded DNA origami to design and construct 3D molecular cages of tetrahedron geometry with inside volume closed by triangular faces. Each edge of the triangular face is approximately 54 nm in dimension. The estimated total external volume and the internal cavity of the triangular pyramid are about 1.8 x 10(-23) and 1.5 x 10(-23) m(3), respectively. Correct formatio...

Compartmentalization of prokaryotic DNA replication.

It becomes now apparent that prokaryotic DNA replication takes place at specific intracellular locations. Early studies indicated that chromosomal DNA replication, as well as plasmid and viral DNA replication, occurs in close association with the bacterial membrane. Moreover, over the last several years, it has been shown that some replication proteins and specific DNA sequences are localized t...

Crystal templating through liquid-liquid phase separation.

Controlled induction of crystal nucleation is a highly desirable but elusive goal. Attempts to speed up crystallization, such as high super saturation or working near a liquid-liquid critical point, always led to irregular and uncontrollable crystal growth. Here, we show that under highly nonequilibrium conditions of spinodal decomposition, water crystals grow as thin wires in a template-less f...

Aqueous Phase Separation as a Possible Route to Compartmentalization of Biological Molecules

How could the incredible complexity of modern cells evolve from something simple enough to have appeared in a primordial soup? This enduring question has sparked the interest of researchers since Darwin first considered his theory of natural selection. Organic molecules, even potentially functional molecules including peptides and nucleotides, can be produced abiotically. Amphiphiles such as su...

Protocell design through modular compartmentalization.

De novo synthetic biological design has the potential to significantly impact upon applications such as energy generation and nanofabrication. Current designs for constructing organisms from component parts are typically limited in scope, as they utilize a cut-and-paste ideology to create simple stepwise engineered protein-signalling pathways. We propose the addition of a new design element tha...
